PROBLEM TO BE SOLVED: To provide a preparation method of a small-granulated HBs antigen capable of obtaining the small-granulated HBs antigen retaining antigen activity. SOLUTION: The preparation method comprises a process of small-granulating an HBs antigen by at least one selected from a group consisting of a surfactant, a reducing agent and a protein-modifying agent; and a process of reacting protein disulfide isomerase with the HBs antigen being subjected to the above small-granulation process. The above process of the small-granulating treatment and the process of the reaction are simultaneously carried out.
