PURPOSE: Provided is an expression vector for manufacturing recombinant Adenovirus which kills cancer cells or inhibits the growth of the cells and is thus useful for anticancer treatments. CONSTITUTION: An expression vector for manufacturing recombinant Adenovirus is provided by the following steps of: i) cutting plasmid pX1 inserted with BamH1 segment having TK gene of HSV1, with restriction enzymes Bgl II and EcoRI to obtain 1.3kb TK gene and 2.3kb DNA segment; ii) inserting the DNA segment into BamH1 and EcoR1 site of plasmid pCMV having 1.16kb CMV promoter to obtain pCMV/TK plasmid; iii) cutting the plasmid with restriction enzyme Xbal to obtain 3.7kb segment; and iv) inserting the 3.7kb segment into Xbal site of Adenovirus transfer plasmid pXCX2 to manufacture expression vector pX/CMV/TK.
