PROBLEM TO BE SOLVED: To easily and quickly obtain a long-chain double-stranded DNA by applying a DNA polymerase to a plurality of template DNAs having at its terminal end a complimentary binding range where the double-stranded DNA can be wholly and continuously formed. SOLUTION: This method for easily and quickly synthesizing a long-chain double-stranded DNA by using >=3 kinds of template single-stranded DNAs is conducted by applying a DNA polymerase to the >=3 kinds of template single- stranded DNAs in the presence of a primer DNA directed to complimentarily anneal a 3'-terminal end side when the 3'-terminal end projects toward the terminal end of the template single-stranded DNA after interlinked, wherein the single-stranded DNAs have a mutually complementary binding range at its at least one of 3'-terminal end and 5'-terminal end, wherein the double- stranded DNA can be wholly and continuously formed by annealing in the binding range, and whereby the long-chain double-stranded DNA having the corresponding whole length is synthetically obtained.
