PROBLEM TO BE SOLVED: To provide a method for synthesizing a dipeptide (an aspartylphenylalanine lower alkyl ester) in a high substrate concentration by using a hydrolytic enzyme (a protease). SOLUTION: In enzymatically producing a dipeptide by condensing an N- substituted aspartic acid (A) with a phenylalanine lower alkyl ester (B) in the presence of a protease, a solution obtained by extracting a substrate B from an aqueous phase to an organic solvent phase and concentrating the organic solvent phase into a concentration to make the concentration of the substrate B equal to or higher than the solubility of the substrate B in water is used as the substrate B. The above substrate A is added to the solution to give a highly concentrated substrate solution, which is used. |
