The present invention relates to methods of assaying for an enzyme that cleaves or joins nucleic acids comprising at least one electrochemiluminiscent label with a sample which may contain a nucleic acid-cleaving or joining enzyme, incubating under conditions that allow cleavage, or joining or hybridizing of said nucleic acid, and measuring the amount of cleaved nucleic acid, and also methods of measuring inhibitors of enzymes that cleave or join nucleic acids.
