(A) Urokinase (I) is isolated from urine by (1) obtaining a ppte. contng. (I), eluting with acid or with aq. 6,9-diamino-2-ethoxyacridine (II) and then recovering (I) from the eluate; (2) contacting aq. soln. of (I) with Ca phosphate gel (or water-insol. alkaline earth metal phosphate) having (II) adsorbed thereon, then eluting when most of pyrogens remain on gel; (3) absorption with carboxymethylcellulose (CMC) from soln. contng. at least 500 CTA units/ml., then eluting when most of pyrogens remain absorbed. Approp. combinations of (1), (2) and (3) used as desired. (B) Human (I) in cryst. homogeneous form. (I) obtd. in good yield and high purity; (I) is used in fibrinolytic treatment of thromboembolic diseases. |
