A pretreatment method for blood analysis comprising the step of selectively labeling leukocytes by treating blood sample with an aqueous solution comprising at least one surfactant selected from the group consisting of a cationic surfactant and an amphoteric surfactant, and with a labeling substance wherein; the aqueous solution of the surfactant is used at a concentration not to damage the whole cell membrane of leukocytes but sufficient to damage a part of the cell membrane; the labeling substance is one of being capable of passing through the damaged cell membrane and combining with an component contained in the leukocytes; and the treatment is performed at pH 3.0 to 11.0.elec
