Methods and compositions for modulating transcription by use of antibodies to transcription factors

Herein described are methods and compositions for inhibiting the activity of a transcription factor in a eukaryotic cell. The methods include introducing into a cell an isolated expressible nucleic acid encoding a nuclear trophic antibody which specifically binds to a transcription factor and inhibits the transcription activity of the transcription factor. The nucleic acid can encode heavy and light chain components of said antibody or a single chain antibody. Also described are methods wherein the nucleic acid is introduced into a germline of a non-human animal to produce a transgenic animal having cells substantially lacking the transcription activity of said transcription factor. The nucleic acid can also be introduced into cells infected with a virus encoding a transcription factor required by said virus to inhibit the transcription activity of said transcription factor. Antibodies to transcription factors and methods of making same are also described.
