PURPOSE: Ura5 gene base sequence coding O rotate phosphoribosyl Transferase(OPRase) is provided to help raising the confidence of transformation. CONSTITUTION: Ura5 gene base sequence of the mold is amplified by Polymerization Chain Reaction to separate Ura5 DNA. Using this DNA segment as a probe, perform southern analysis with the entire digested M. anisopliae DNA and identify that the above separated DNA segment is came from M. anisopliae. Then the entire genome gene is separated from M. anisopliae, cloned and sequenced.
