PROBLEM TO BE SOLVED: To obtain peptide (or its derivative) in high efficiency by carrying out the peptide synthesizing reaction of amino acid (or their derivative) raw material and enzyme which expresses enzymatic activity in an organic solvent, in water/the organic solvent mixed phase and collecting the product form water phase. SOLUTION: The peptide usable for a precursor or the like of kyotorphin or the like for a analgesic or the like in high efficiency by carrying out the peptide synthesis reaction of the enzyme E expressing enzymatic activity in the organic solvent and covered with a surface activity, and amino acids or their derivatives S comprising amino group protected tyrosine and carboxyl group protected arginine or the like adding to the water/organic solvent mixed system, carrying out the peptide synthesis reaction in organic solvent phase by the enzyme, collecting after transferring the synthesized peptide or the derivatives P into the water phase.
