PURPOSE: A nucleic acid molecule encoding enzymes taking part in starch synthesis in plants, a vector containing the nucleic acid molecules and host cells which have been transformed with the nucleic acid molecules, transformed plant cells and regenerate plants are provided. CONSTITUTION: Polyclone antibody (anti-SS1) was prepared by using a peptide of SEQ ID No:3, and cDNA library encoding the soluble maize starch synthases was screened and E.coli clone was obtained by using the anti-SS1 antibody. Plasmid pSSS1 was separated from the E.coli clone. The nucleic acid has a sequence of SEQ ID No:1 encodes the protein comprising amino acids of SEQ ID No:2. The protein is an enzyme involving a novel isoform of soluble maize starch synthases. Plasmid vector pUBI-bar-aMASY was prepared by using vector pUBIbar (See Fig. 1) and comprise a bar marker gene consisting of ubiquinone promoter, intron of maize, transcription-terminating signal of nopaline synthase gene of A. tumefaciens, coding area of bialaphos-resistant gene of Streptomyces hygroscopicus, and a terminator of CaMV connected with 35S promoter and bar gene (See Fig. 2). The vector pUBI-bar-aMASY was introduced into protoplasm to obtain 408 phosphinotricin-resistant clone and clone 6 among clones was regenerated to a plant.
