摘要 PROBLEM TO BE SOLVED: To obtain an integrated host capable of recovering a DNA converted into a plasmid by directly integrating a target DNA of the complete length having DNA regions as footholds at both of its terminals into bacterial genome or the like without using any vector. SOLUTION: This host integrated with a DNA is obtained by directly integrating a target DNA of the complete length having DNA regions as footholds at both of its terminals into a genome of e.g. bacteria, such as Bacillus subtilis, without using any vector and repeatedly conducting this integration manipulation and is usable e.g. for gene modified plants and gene therapy. This system is capable of recovering a DNA portion, as a circular plasmid, integrated into a genome by surely cloning a DNA having a large size exceeding 50-100 kb and subjecting the genome to cleavage treatment and capable of creating bacteria afforded with new properties enabling e.g. substance production and environmental clean-up and also is usable e.g. for gene modified plants and gene therapy.
