A new diagnostic agent for the detection of leukocytes in body fluids contains an alpha -hydroxyvaleric acid ester, an optional activator and a diazonium salt. A diagnostic agent contains, as the substrate, an alpha -hydroxyvaleric acid ester of formula (I): R = a group that can be detected as R-OH once the ester has been cleaved (RO is e.g. indoxyl, thioindoxyl, phenyl or naphthol); and R' = a protecting group (e.g. an arylsulfonyl or alkylsulfonyl group). An Independent claim is also included for the following: (1) O-(4'-toluolsulfonyl)- alpha -hydroxyvaleric acid-3-indoxyl ester (Ia); and (2) preparation of (Ia) by reacting alpha -hydroxyvaleric acid ethyl ester with tosyl chloride, hydrolyzing the resulting ethyl ester, reacting the resulting carboxylic acid with thionyl chloride to give an acid chloride, and reacting with an indoxyl moiety.
