PROBLEM TO BE SOLVED: To measure the number of cells in leucocyte by using a nitrogen- containing heterocyclic compound having a specific pH value as a dissolving reagent used in the cell measurement and analyzation of leucocyte. SOLUTION: A dissolving reagent includes an effective amount of anticoagulant. As the examples of the anticoagulant, heparin, noticeably, citric acid ion, and particularly EDTA are included. A mixture of 0.15 M of pyrolidone chloride, 0.04 M of boric acid, 0.0001 M of EDTA and 0.005 M of HEPES is used at approximately 7.0 pH under the priority condition for the total use. For the labeling of the leucocyte cells, the operation can be executed in the absence of or in the presence of a monoclonal antibody. The antibody is bonded to a fluorescent compound. The dissolving reagent completes the dissolution by contacting 0.1 mol of blood sample treated by the anticoagulant, and incubated in advance with a monoclonal antibody, or a monoclonal antibody mixture combined with a fluorescent marker, with 2 ml of the dissolving reagent heated by 37 deg.C, and leaving and cooling the same as it is for 10 minutes. |
