PROBLEM TO BE SOLVED: To provide a method for quickly counting Lactobacillus heterohiochii without needing any expensive instruments in order to improve the quality of Japanese sake, by staining the colonies derived from Lactobacillus heterohiochii obtained under a specific condition by using a basic dyestuff and then counting the colonies. SOLUTION: This method for counting the number of Lactobacillus heterohiochii in a sample is to conduct a filtration, collection and cultivation by using a membrane filter and count the resultant colonies, wherein the cultivation is carried out in an anaerobic condition for a predetermined time by using a solid culture medium for Lactobacillus heterochiochii comprising at least one of agar, gelatin and carrageenan, wherein the colonies are obtained by treating the outer surface of the membrane filter with an organic solution of a basic dyestuff and air-drying it, and wherein the number of the stained colonies is counted by microcope or magnifier. |
