PROBLEM TO BE SOLVED: To provide a method for determining a whole sequence by directly sequencing fragments from a long DNA by restriction enzymes from PCR products. SOLUTION: This is a method for preparing a DNA sample for determining a base sequence of a DNA, which comprises the step of screening and amplifying a specific fragment from DNA fragments using at least one pair(s) of primers consisting of a first primer selected from a first set of primers and a second primer selected from a second set of primers, wherein the first primer set consists of a plurality of primers having base sequences complementary to base sequences recognized by a first restriction enzyme, the second primer set consists of a plurality of primers having base sequences complementary to base sequence recognized by a second restriction enzyme and the second restriction enzyme recognizes a base sequence different from the base sequence recognized by the first restriction enzyme.
