A method of chromatography in which a chromatograph having a circulation system comprising unit packing layers (1) to (4) endlessly connected in series is used for separating three ingredients A, B, and C whose affinity for the packing is in the order of CBA, which comprises a step (1) as the first stage in which the following operations 24143 are conducted and a subsequent step (2) as the second to fourth stages which comprises three repetitions of the following operations 243, the liquid introduction point and the liquid discharge point each being shifted by one unit layer toward the downstream side in the circulation system after every stage: 1 the circulation system is interrupted, and all the ingredient B fraction is discharged from the upstream side of the interruption point while supplying a feed fluid f to the unit packing layer on the immediately downstream side of the interruption point and further supplying an eluent D to the circulation system; 2 the ingredient A fraction is partly discharged while supplying the eluent D to the circulation system; 3 the ingredient C fraction is partly discharged while supplying the eluent D to the circulation system; and 4 liquid circulation is conducted through the circulation system without conducting liquid supply to the system or liquid discharge therefrom.
