METHODS AND COMPOSITIONS FOR NUCLEIC ACID DELIVERY

Nucleic acid delivery compositions, methods for delivery of nucleic acid in vivo to an animal or in vitro to target cells and cellular populations, such as lymphoid cells, having internalized therein the inventive nucleic acid delivery compositions are provided. In one form of the invention, a nucleic acid delivery composition includes polymeric microparticle matrices, especially polysaccharide microparticles such as alginate microparticles, and a nucleic acid embedded in the microparticles. In other embodiments, the nucleic acid delivery composition further includes a virus composition, such as an adenovirus composition. The polymeric microparticles may be coated with a cationic polymer and are, in one inventive form, microspherical. Methods of in vivo delivery of a nucleic acid to a target cell are also described. The methods include administering to an animal an effective amount of a nucleic acid delivery composition as described above. Methods of in vitro delivery of nucleic acid to target cells that include incubating the cells with the nucleic acid delivery compositions of the present invention are also provided.
