PROBLEM TO BE SOLVED: To obtain a protein composition containing a cooperative interferon, etc., by binding a reactive aldehyde group-containing water-soluble polymer to anα-amino group such as a protein or the like under a reductive alkylation reaction condition and then separating the reaction product. SOLUTION: In this method for binding a water-soluble polymer containing a single reactive aldehyde group to a protein or its analog, the protein part is reacted with a water-soluble polymer part of a poly(N-vinylpyrrolidone), a polyethylene glycol, a polyvinyl alcohol or the like under reductive alkylation conditions using sodium boron hydride or the like at an acidic pH sufficient to selectively activate theα-amino group of the amino end of the protein part to selectively bond the water-soluble polymer to theα-amino group and the reaction product is separated from the unreacted part to give the objective chemically modified protein composition such as an N-terminal chemically modified G-CSF composition, a chemically modified cooperative interferon, etc. |
