PURPOSE:To enable the assay of creatinine and creatine in a body liquor accu rately with a high specificity at a high sensitivity without being affected by any coexisting substance, by using four enzymes - CRN, CR, SOX and peroxidase. CONSTITUTION:One aliquot in a sample is mixed with three enzymes, namely, creatinine amidohydrolase, creatine amidinohydrolase and sarcosine oxydase to act upon one another so that creatinine and creatine in the sample is converted into hydrogen peroxide, formaldehyde and glycine. The other aliquot of the sample is mixed with two enzymes such as creatine amidinohydrolase and sarcosine oxidase to act upon one another so that creatine in the sample is converted into hydrogen peroxide, formaldehyde and glycine. The hydrogen peroxide generated as mentioned above is made to react with color forming reagent for assaying hydrogen peroxide in the presence of peroxydase to measure the color formed body generated. Thus, the total of creatinine and creatine as mentioned above and the amount of creatine are determined and the amount of creatine is subtracted from the total amount to calculate the amount of creatinine. |
