Disclosed is a method of making a ribonucleic acid transcript of a deoxyribonucleic acid sequence, the method comprising: providing the deoxyribonucleic acid sequence to be transcribed; and treating the sequence under suitable conditions with a reaction mix comprising an RNA polymerase and a molecule in accordance with the structure shown in the figure, where X<1> may be O, S, N-alkyl, N<+>-dialkyl or N-benzyl; and wherein X<2> may be triphosphate (P3O9)<4->, diphosphate (P2O6)<3->, thiotriphosphate (P3O8S)<4-> or an analogue thereof, together with a method of isolating an RNA molecule of interest, and a method of determining the position in an RNA sequence of one or more degenerate base analogues.
