NOVEL MONOCLONAL ANTIBODY, AND IMMUNOLOGICAL ANALYSIS OF C-SKI PROTEIN AS CANCER GENE PRODUCT

PROBLEM TO BE SOLVED: To obtain a novel monclonal antibody composed of a monoclonal antibody or its fragment reactive with human c-Ski protein, and useful for, e.g. diagnosis of, e.g. the presence or absence of canceration by immunological analysis of the human c-Ski protein as the cancer gene product. SOLUTION: This antibody is composed of a monoclonal antibody or its fragment reactive with human c-Ski protein, and useful for, e.g. immunological analysis of c-Ski protein, a product of c-Ski gene as one of cellular cancer genes, and also for diagnosis of presence or absence of canceration and its extent, when it is present. This monoclonal antibody is obtained by introducing a manifestation vector containing a fragment of human cancer gene of c-ski- CDNA treated with a restriction enzyme into Escherichia coli, to prepare c-Ski protein, administering the protein into a mouse's abdominal cavity after emulsifying it with adjuvant, taking out its spleen cells after the protein is immunized, to fuse them with mouse's myeloma cells, culturing them in a HAT medium to produce the hybridoma, and cloning and then culturing the hybridoma.
