PROBLEM TO BE SOLVED: To obtain a reagent composition for measuring chloride ion in serum, useful as a clinical examination, etc., and having high measuring accuracy by formulating an inactivated typeα-amylase with a chelating agent and a maltooligosaccharide derivative as a substrate. SOLUTION: (A) An inactivated typeα-amylase (e.g. bovine pancreas-derivedα-amylase) is formulated with (B) a chelating agent (e.g. ethylene diamine tetraacetate) and (C) a maltooligosaccharide represented by the formula [R1 and R2 are eachβ-galactopyranosyl group or H; R3 is 2-chloro-4-nitrophenol group; (n) is an integer of 0-2] (e.g. 2-chloro-4-nitrophenyl-α-maltoside) as a substrate to provide the objective reagent composition capable of measuring chloride ion in high sensitivity and accuracy without using a following enzyme. Furthermore, the reagent composition is useful for diagnosis of chloropenia such as hypotonic dehydration, glucocorticoid supernumerary symptom or respiratory acidosis and hyperchloromia such as hypertonic dehydration or uriniferous tubular acidosis.
