Selecting genes that express mutant proteins of high stability, especially enzymes and binding proteins

A method for selecting a gene (A) that encodes a protein which inhibits the replication of (A) is new. The method uses an infectious replicable gene package (IRG) for coupling the ability to replicate to the stability of a protein (PR) encoded by (A). Independent claims are also included for the following: (1) a method for selecting genes that express mutant PR with increased stability to proteolysis, using IRG; and (2) a kit for selecting genes comprising a vector that can be packaged in IRG and includes: (i) a cloning site for insertion of DNA into a region encoding a protein that is essential for infectivity of IRG; and (ii) a modification which suppresses infection of a host by IRG when this is subjected to partial proteolysis.
