PROBLEM TO BE SOLVED: To obtain a new luciferase having an amino acid sequence of a firefly luciferase having mutation at a specific cite, further having resistance to a surfactant, useful for measurement or the like of presence or absence of cells of microorganisms or the like in a sample, or the number of cells, and usable for measurement of ATP in the cell. SOLUTION: This new luciferase comprises a polypeptide having an amino acid sequence of the formula, or a polypeptide having the amino acid sequence of the formula having the insertion, deficiency or substitution of one or plural amino acids, and further having surfactant resistance and luciferase activities, and has resistance to the surfactant. The luciferase is useful for measurement or the like of presence or absence of cells in a sample, or the number of cells in a field such as food sanitation, bio, clinical examination, medical, extrapure water and environmental fields, and usable for measurement or the like of ATP in the cell. The enzyme is obtained by inducing a mutation to a gene so that glutamic acid at the same position as 490-position of the luciferase of Luciola cruciata or the like may be substituted with other amino acid by a cite-specific mutation-inducing method, and allowing the obtained gene to be expressed. |
