PROBLEM TO BE SOLVED: To obtain a new monoclonal antibody capable of specifically recognizing human N-acetylglucosamine transferase-V (human GnT-V) and useful for immunological GnT-V measurement expected to contribute to the diagnosis of malignancy of cancer. SOLUTION: A monoclonal antibody specifically recognizing human GnT-V. The antibody is obtained by immunizing an animal with a polypeptide of the formula corresponding to the region of the 431-741 amino acids in the human GnT-V as an immunogen, collecting antibody-producing cells originated from the spleen of the immunized animal, fusing the cells to tumor cells to form a hybridoma [for example, hybridoma GNTV13F4 (FERM P-16554), and subsequently culturing the hybridoma to produce the antibody. The above polypeptide is also obtained, for example, by proliferating the GnT-V region by PCR method with reference to the known base sequence of human GnT-V, cleaving the proliferation product with restriction enzymes to obtain a DNA fragment having a region encoding the polypeptide, introducing the DNA fragment into a vector, transforming a host with the vector and subsequently culturing the transformant to produce the polypeptide. |
