摘要 Chimaeric gene formed by DNA sequences which code for specific antigens to four proteins of L. infantum, applicable in the serological diagnosis of canine leishmaniosis and protein obtained. Chimaeric gene formed by the partial DNA sequences which code for the specific antigens for four Leishmania infantum proteins (LiP2a, LiP2b, LiPH2A, LiP0) and the protein obtained which has application in the diagnosis of leishmaniasis. The cloning strategies for a chimaeric gene and the intermediate products generated during the process of forming the gene are described. Use of the partial clone rLiPP0-Ct-Q as a vector is indicated, and the partial DNA fragments of the LiP2a, LiP2b and LiH2A proteins are added sequentially to this vector in the 5' direction using appropriate restriction enzymes. The steps followed in determining the correct orientation of the DNA inserts in the final gene and the intermediate protein products are described. The final polypeptide generated has a molecular mass of 38K and an isoelectric point of 7.37. The nucleotide sequence of the chimaeric gene and the deduced amino acid sequence of the synthesised protein are described. <IMAGE>
