CULTURE METHOD FOR HUMAN STEM CELL-CONTAINING COMPOSITION AND TRANSFORMATION THEREOF

PROBLEM TO BE SOLVED: To efficiently obtain a human dividing stem cell-containing composition that is useful for proliferation, transformation and the like of cultured mammalian cells by culturing human dividing stem cells observed in the human hematopoietic system ex vivo in the presence of hematopoietic stem cell grow factor. SOLUTION: Human dividing stem cells and/or human dividing hematopoietic precursor cells observed in bone marrow are continuously or periodically exchanged or perfused in a liquid culture medium, nutrients are supplied under the physiologically acceptable conditions and an effective amount of hematopoietic growth factors are added to culture these cells ex vivo thereby producing the objective human dividing stem cell composition that is useful for proliferation, transformation and the like of cultured mammalian cells. The hematopoietic cell growth factor to be used here is, for example, erythropoietin(Epo), interleukin-3(IL-3), granulocyte macrophage colony stimulation factor(GM-CSF), granulocyte colony stimulation factor(G-CSF), IL-6, IL-7, IL-11 or stem cell factor and the like.
