PROBLEM TO BE SOLVED: To obtain a new vector comprising a nucleic acid having a specific minimum and maximum size, capable of carrying out cloning of DNA fragment which is too large to clone with plasmid and too small to clone with cosmid and suitable for cloning, etc., of DNA proliferation product, etc. SOLUTION: This new cosmid vector comprises Escherichia coli ColE1 duplication product, ampicillin-resistant gene, multiple cloning cassette, cos part for in vitro packaging in λ packaging extract and a fragment from genome of bacteriophage λ and has 15 kb minimum size and 35 kb maximum size and is capable of cloning DNA fragment having a certain size category which is too large to clone with plasmid and too small to clone with cosmid and suitable for cloning of DNA proliferation product, etc. The cosmid vector is obtained by binding cos sequence of bacteriophage, replication origin of prokaryote and gene, etc., for selection in antibiotic culture medium. |
