The invention relates to a method for producing a purified recombinant TSH receptor preparation (rTSHR preparation). According to said method, a recombinant fusion-TSH-receptor is produced with an additional amino acid sequence inserted between the amino acid sequence of the functional TSH-receptor and a peptide residue which can be bonded to a solid phase. Said additional amino acid sequence can be selectively split off from the amino acid sequence of the functional TSH-receptor using a suitable protease, e.g., factor Xa. The fusion receptor is bonded to a suitable solid phase, washed to remove contaminant constituents and optionally marked in bonded form. The fusion receptor which is bonded to the solid phase is treated with protease to proteolytically split off the amino acid sequence of the functional TSH-receptor from the solid phase-bound residue of the fusion receptor and the resulting purified rTSHR preparation is prepared. The purified or purified and marked human rTSHR preparation can be used for oral toleration and for producing a novel immunoprecipitation assay for analysing anti-TSHR auto-antibodies.
