The present invention relates to the use of topo I and agents which enhance topo I activity to improve the efficiencies of retroviral-based gene therapies and reverse-transcriptase-based molecular biology techniques. In accordance with the present invention, topo I and agents which enhance topo I may be used to improve integration efficiencies of retroviral vectors and have utility in improving retroviral vector-based gene delivery and gene therapy techniques.
