PROBLEM TO BE SOLVED: To obtain the subject new enzyme containing a specific amino acid sequence, having an enzyme activity of norcoclaurine 6-O-methyltransferase and useful for the production, etc., of various alkaloids derived from coclaurine or reticulin. SOLUTION: This new polypeptide contains an amino acid sequence described in the formula or an amino acid sequence in which one or more amino acids are added, deleted or replaced in the amino acid sequence and having an enzyme activity of norcoclaurine 6-O-methyltransferase(6-OMT) and is useful for the efficient production, etc., of coclaurine, reticulin and/or various alkaloids which are plant secondary metabolites biosynthesized from the compounds. The enzyme is obtained by preparing a DNA probe based on an internal amino acid sequence of the 6-OMT isolated from a cultured cell of Coptis japonica Makino, searching for a DNA library derived from a cultured cell of the Coptis japonica Makino using the prepared DNA probe, integrating the resultant gene into a vector and expressing the gene in a host cell. |
