PURPOSE:To enable detection with an excellent proliferation ratio by blending peptone with NaCl, a yeast extract, glycerol, KH2PO4, sodium pyruvate, KNO3, bovine bile powder and agar in prescribed amounts. CONSTITUTION:A culture medium is obtained by blending 1.0-2.5wt.% peptone with 0.25-0.5wt.% NaCl, 0.4-0.6wt.% yeast extract, 0.5-1.5wt.% glycerol, 0.2-0.5wt.% KH2PO4, 0.05-5.0wt.% sodium pyruvate, 0.05-1.0wt.% KNO3, 1.0-2.0wt.% bovine bile powder, 0.1-0.3wt.% agar and, as necessary, 10<-4> to 10<-2>M 4-methyl-umbelliferyl-beta-galactoside or 4-methyl-umbelliferyl-beta-d-glucuronide. The resultant culture medium is then regulated so as to provide <3 min generation time and >=0.35 proliferation ratio without any lag phase. Thereby, the objective culture medium for detecting the coliform group is produced. |
