PROBLEM TO BE SOLVED: To provide a method for isolating a genome DNA usable for determination, etc. of gene type by amplifying the genome DNA of each specimen by using a primer with mild selectivity, mixing the amplified products to hybridize, and amplifying the formed double strand DNA. SOLUTION: This method for amplifying genome DNA regions with different base sequence among specimens to be tested comprises carrying out a polymerase chain reaction by using a primer having a mild selectivity, and the genome DNA of each specimen as a template, mixing the DNA of each specimen amplified by the polymerase chain reaction, modifying the mixture, hybridizing the mixture and selectively amplifying the double strand DNA formed by the hybridization and derived from only one specimen by the polymerase chain reaction. By using the method, the genome DNA region useful for a simple and effective method for determining gene types, and having different base sequences among the specimens can be efficiently amplified and isolated. |
