An nCL-3 calpain gene (I) with a defined sequence of 2459 bp given in the specification and its allelic variants, analogues or derivatives, are new. Also claimed are: (1) a gene construct in which (I) is functionally linked to one or more regulatory signals for increasing gene expression; (2) the 640 amino acid gene product of (I); and (3) a method for identifying calpain inhibitors, comprising isolating a calpain encoded by the sequence of (1) from tissues or cells in which the nCL-3 enzyme is expressed, determining the inhibition of the cleavage of an nCL-3 substrate and calpain I and/or II substrates by test substances, and selecting test substances that exhibit an inhibitory action at least against one of the calpains. |
