The invention is directed to compositions and methods for the preparation of a composition containing conditioned medium that enhances the growth of antigen presenting cells in culture. Compositions are specifically useful for the growth and maintenance of dendritic cells, macrophages, and monocytes. Methods comprise culturing a population of cells in a composition containing a conditioned medium prepared according to the methods of the invention. Cultured cells can be further treated with compounds which reduce, suppress or eliminate the growth of undesired cell types, such as T-cells. Cells that proliferate according to these methods comprise enriched populations of dendritic cells, monocytes and macrophages. Such cells are useful in the creation of cellular vaccines and for immunotherapy, and can serve as useful vehicles for gene therapy.
