PROBLEM TO BE SOLVED: To obtain a polynucleotide having on a single strand a molecule, such as ribozyme or antisense RNA molecule, having high therapeutic effects on AIDS, tumors, and the like. SOLUTION: This ribonucleotide has a structure of formula I: 5'-α1 β1 α2 β2 ...αn βn -3', formula II: 5'-β1 α1 β2 α2 ...βn αb -3' or the like [α1 -αn are polyribonucleotides capable of forming complexes with polyribonucleotides γ1 -γn , respectively; β1 -βn are each a polyribonucleotide; (n) is an integer of >=1]. The γ1 -γn can form complexes having ribozyme activities together with the α1 -αn , respectively. The molecule of formula I or formula II is cut at the prescribed positions of the α1 -αn , respectively, in a cell. The cut fragments containing β1 -βn have activities specifically inhibiting the functions of polynucleotides δ1 -δn (existing in RNA originated from HIV or the like) in a cell. The polynucleotide is obtained by ligating a DNA transcribed to the polynucleotide of the present invention to the downstream of a promoter sequence such as T7RNA polymerase and subsequently treating the ligation product in the presence of ATP, CTP, GTP and UTP. |
