FIELD: biotechnology, genetic engineering. SUBSTANCE: method of preparing hirudine HV1 is carried out by transformation of recombinant strain Escherichia coli JM-109 (BP-32266) with secretion vector pMTS HV1 or pMKS HV1 followed by its culturing and isolation of the end product from cultural medium or cell periplasm. Secretion vector has PvuI-PvuII-fragment (size is 1440 base pairs) containing a replication region of plasmid pUC18, trp- or tac-promoter, DNA fragment encoding signal peptide of an alkaline phosphatase, DNA fragment encoding hirudine HV1 combined in site AccI and gene determining the resistance to ampicillin as a genetic marker.
