The present invention relates to compositions and methods for enhancing gene delivery. The invention provides a method for enhancing uptake of an exogenous nucleic acid by cells of a mammalian tissue, wherein a nucleic acid is administered to a mammal in combination with a substance that restricts the efflux of blood from that tissue, such that nucleic acid uptake by the cells is increased relative to uptake in the absence of the substance. The invention also provides compositions for use in the method of the invention.
