PROBLEM TO BE SOLVED: To detect the position data of protein with high sensitivity by forming a fluorescent image of protein high in sharpness and low in background noise, by dyeing electrically migrated protein by a fluorescent dye for dyeing protein, and irradiating dyed protein with exciting light to obtain fluorescence to photoelectrically detect the same. SOLUTION: Protein electrically migrated on a gel is fixed on the gel by using a fixing soln. and dyed by using a fluorescent dye mainly acting on a sodium dodecyl sulfate-protein composite to dye protein, and dyed protein is irradiated with exciting light to generate fluorescence which is, in turn, optically detected to form a fluorescent image of protein. By this method, the fluorescent image high in sharpness and low in background noise is formed and the detection sensitivity of the position data of protein is enhanced. If dyed protein is decolored by using a decoloring soln. after dyeing, sensitivity is further enhanced. As the fixing soln., an acetic acid and methanol or ethanol-containing soln. is designated and the optimum contact time with the fixing soln. is 15 min, the dyeing time of protein is pref. 1-2 hr. |
