AMPLIFICATION AND DETECTION OF NUCLEIC ACID SEQUENCE

PROBLEM TO BE SOLVED: To provide a process for the quick and accurate detection of a specific nucleic acid sequence in a test specimen. SOLUTION: This process for the production of a double-stranded nucleic acid containing a promoter operably bonded to a sequence to be detected comprises (a) the production of an oligonucleotide promoter-primer, (b) the contact of the promoter-primer with a nucleic acid containing the sequence to be detected under a condition to cause the hybridization of the promoter- primer and the nucleic acid sequence to be detected, (c) the production of an extension product complimentary to the objective nucleic acid from the 3' terminal of the promoter-primer, (d) the contact of the product of the step (c) with a substance having 3'-5'-exonuclease activity and (e) the synthesis of an extension product complimentary to the promoter of the promoter-primer from the 3'-terminal of the sequence to be detected.
