ANTI-HBS MONOCLONAL ANTIBODY

PROBLEM TO BE SOLVED: To obtain a safe anti-HBs monoclonal antibody high in binding ability to the surface antigen of hepatitis B, and useful as a diagnostic or a remedy for the hepatitis B. SOLUTION: This anti-HBs monoclonal antibody has the following properties: (1) CDR-3 in a variable range of an H chain has an amino acid sequence of ThrAlaPhePheAsnAlaTyrAspPhe; (2) the antibody includes no EBV(Epstein-Barr Virus); and (3) the antibody binds with at least adr type in human HBs antigen. Fab fragments of the monoclonal antibody containing no EBV can be obtained by cloning each of DNA fragments encoding H-chain and L-chain of the antibody from cells [e.g. E. coli JM 109 (E.coli-HBs-CL4-3)(FERM P-16421)] producing the anti-HBs monoclonal antibody having a high antibody titer, and expressing the cloned DNA fragments in the E. coli.
