PURPOSE:To obtain the title phosphatase resistant to inhibition by inorganic phosphoric acid as a degradation product from enzymatic reaction, thus useful as a labelled enzyme for the enzyme immunoassay by using microorganisms short in culture period, classified as thermophilic Bacillus. CONSTITUTION:Microorganisms classified as thermophilic Bacillus and capable of producing alkali phosphatase resistant to inhibition by inorganic phosphoric acid [pref. Bacillus stearothermophilus TB-443 (FERM P-10522)] is put to culture, and said alkali phosphatase is produced and accumulated in the culture solution, and then collected, thus obtaining the objective phosphatase resistant to inhibition by inorganic phosphoric acid. For the culture conditions, it is suggested that the temperature be 45-55 deg.C and the time 6-24hr. |
