<p>The present invention provides a method of characterizing an isolate of a human immunodeficiency virus as CCR5 dependent or CXCR4 dependent comprising: a) contacting the isolate of human immunodeficiency virus with mitogen-stimulated peripheral blood mononuclear cells under conditions that would permit replication of the virus; b) contacting some of the peripheral blood mononuclear cells from step (a) with an amount of interleukin 4; c) measuring the amount of viral replication in the cells of step (b); d) measuring the amount of viral replication in the peripheral blood mononuclear cells of step (a); and e) comparing the amount of viral replication measured in step (c) with the amount of viral replication measured in step (d), whereby a greater amount of viral replication in the cells of step (c) indicates a CXCR4 dependent isolate of human immunodeficiency virus and a lesser amount of viral replication in the cells of step c) indicates a CCR5 dependent isolate of human immunodeficiency virus.</p> |
