Methods and compositions for generating chemiluminescence on reaction with a peroxidase enzyme are provided as well as novel compounds useful therein. The compounds comprise a C-C double bond substituted at one carbon with two oxygen or sulfur atom-containing groups. The compositions comprise the double bond containing compound, a peroxide and optionally a peroxidase activity enhancing substance in an aqueous solution. The compositions can additionally comprise a nonionic surfactant or a cationic surfactant or both to improve detection sensitivity or the peroxidase. The novel chemiluminescent methods and compositions are useful in assays for peroxidase enzymes and in assays employing enzyme-labeled specific binding pairs.
