PROBLEM TO BE SOLVED: To provide a method for determining the base sequence of a DNA, enabling to produce a long chain transcript product and obtain sequence data little in the irregularity of signals from a labeled deoxyribonucleotide in improved accuracy. SOLUTION: This method for determining the base sequence of a DNA comprises reacting a ribonucleotide-5'-triphosphate with a 3'-dNTP derivative in the presence of a DNA fragment and a variant RNA polymerase modified so as to improve an ability for taking the 3'-dNTP derivative, separating the obtained nucleic acid transcript product and subsequently reading the sequence of the nucleic acid from the separated fraction. The DNA fragment contains a promoter sequence for the RNA polymerase. |
