FIELD: molecular biology, genetic engineering, biochemistry. SUBSTANCE: gene encoding streptokinase is subjected for amplification using PCR and recombined with procaryotic expressing vector - plasmid PLY-4. Cells of E. coli pSTE-SK-I are transformed to express gene SK by temperature effect. Obtained bacteria are fermented and disrupted by high pressure. Inclusion bodies are collected and the isolated r-sk is purified for two stages involving ionic exchange and gel-filtration. Purity and yield of r-sk are high at low cost.
