PROBLEM TO BE SOLVED: To obtain various and preferable variants D-amino acid oxidase, by preparing D-amino acid oxidase useful for producing an α-ketoacid, resolving a racemic mixture of amino acids, analysis and determination of amino acid, etc., using a fungus belonging to the genus Rhodosporidium as an isolation source. SOLUTION: This isolated amino acid oxidase is derived from a fungus of the genus Rhodosporidium and comprises, for example, am amino acid sequence of the formula. A D-amino acid oxidase isolated from Rhodosporidium toruloides ATCC 10,788 has 37 kDa (SDS/pAGE) and optimum pH 9.0 is stable in the pH range of 7.0-8.0 and has 35 deg.C optimum temperature and activity at 30-50 deg.C exceeding 70% the maximum activity. A transformant DH5 α strain by expression plasmid pDAO-23 having D-amino acid oxidase gene derived from R. Toruloides has obtained deposition number 98,485 from ATCC. |
