摘要 The assembly to measure the residual fluorescence in hydronicotinamide adenine dinucleotide (NADH), in cell cultures, has an Ulbricht globe photometer (1) with an entry (122) and exit (123) opening on the equator of a perfusion chamber (13) which holds the cell culture (1332). The entry opening (122) has a system to excite the residual NADH fluorescence with pulsed light at the cell culture (1332) within the perfusion chamber (13). A pulse detector (4) is at the exit opening (122) to register the emitted fluorescent light from the cell culture (1332). The pulse detector (4) is a photo receiver (42) behind an edge filter (41) and followed in series by a gate circuit (431), pulse amplifier (43), sample and hold circuit (44) and an analog/digital converter (45). The pulse detector is a polychromator with a following charge coupled device (CCD) receiver and CCD control electronics. A pulse laser (2) gives the pulsed excitation light, and pref. a nitrogen laser. The perfusion chamber (13) is cylindrical, with a dia. larger than its height. The end sides of the perfusion chamber are formed by glass discs, transparent to ultra violet light, with a coating on the outer surfaces to reduce reflection. The covering glass (133) of the perfusion chamber (13) can be removed, and used as the culture plate for the cell cultures (1332) under study.
