The invention disclosed is a staphylokinase (Sak)-producing Staphylococcus aureus SL1.063, a novel recombinant staphylokinase and their high expression engineered strain. A gene coding for Sak was obtained by PCR amplification with chromosomal DNA of Sak-producing Staphylococcus aureus as a template. The nucleotide sequence of the gene and the amino acids they encoded was different from those of the Sak gene which had been reported. And the recombinant DNA fragment was obtained by amplification using a primer which had a particular nucleotide sequence, purified, identified, cut with enzymes, ligated and transformed into a host cell to construct a Sak-high expressing engineered strain, the expression product acted as plasminogen activator and had very high hydrolytic activity to human fibrin. It cold treats thrombembolia disease such as myocardial infarction, cerebral, pulmonary thrombus and the like. |
